Arcturus XT Laser Capture Microdissection System
This instrument allows the isolation of homogeneous populations/regions for analyses of DNA, RNA and protein. Supports microdissection of single cells, groups of cells or defined regions in a variety of preparations. This system consists of a motorized Nikon TE 2000U inverted microscope, an infrared laser and an ultraviolet laser for laser capture and cutting. The microscope is equipped for bright field, phase and blue, green and red fluorescence microscopy. A second high resolution digital camera is installed for publication quality image capture.
Laser capture microdissection is a technique for isolating a specific population of cells from a heterogenous tissue section or cytological specimen by using a thermolabile polymer membrane and IR laser that makes polymer sticky. Precise isolation can also be improved by cutting the sample with UV laser. Samples from formalin-fixed paraffin embedded tissues to live cells can be dissected. The process consist of (1) visualization of the cells with a microscope, (2) laser transfer to a polymer membrane for formation of a polymer-cell composite, and (3) removal of the cells from the heterogenous environment. The isolated samples can be processed for molecular components (DNA, RNA, and proteins) to obtain molecular profile of specific cell populations. Various kits for preparation samples for dissection, staining, and purification of macromoleculs are available from the Molecular Devices to maximize the efficiency of microdissection and purification.
