Research Interests

  • Our goal is to understand the spectrum and biology of human disease associated with immune dysregulation in primary immunodeficiencies as well as to define strategies for the early detection of susceptible individuals and novel approaches for treatment. In particular, we are focusing on targeted treatment strategies for autoimmune complications, such as cytopenias. As a prototype of this condition, we primarily study patients and murine models with partial defects in the genes responsible for maturation of antibody genes, namely the recombination activating genes 1 and 2 (RAG1/RAG2). The enzymes encoded by these genes initiate V(D)J recombination and allow pre-immune diversification of the T- and B- cell repertoires. Drawing on observations in patients as well as mouse models of RAG deficiency, we have described impaired central and peripheral checkpoints in B cell tolerance and published extensively on the phenomenon of immune dysregulation and altered T- and B cell repertoire.
