Research Interests

  • Basic research: My basic research focuses on the regulation of cell growth and development in epithelial ovarian cancer (EOC), the most frequent and lethal gynecologic malignancy. Using a gene discovery approach, a number of molecules that regulate blood vessel formation (angiogenesis) and tissue architecture have been identified as significantly up-regulated or down-regulated in the main tissue component of EOC. Development of tumor structures is also being re-constructed and investigated in vitro utilizing recombinant cultures of EOC cells, blood vessel lining cells and fibroblasts grown in extracellular matrix.
  • Translational research: My laboratory has shown that EOC development is regulated in a “Yin and Yang” fashion by factors that promote or suppress formation and growth of blood vessels, structures integral to tumor development. Some of these factors are dramatically elevated in the cyst fluid of EOCs consistent with the hypothesis that cytokines may play an important role in ovarian cancer related-angiogenesis and tumor progression. Translating these results to the bedside, the laboratory has shown that seriousness of clinical status correlates with urinary angiostatin, an angiogenesis inhibitor that independently discriminates EOC patients from volunteer controls and patients with benign ovarian masses. A second potential biomarker under current study is the protein product of the High Motility Group A2 gene, an architectural transcription factor that is over-expressed in high grade serous ovarian and tubal carcinomas. Discovery and validation of urinary biomarkers of EOC continue to be main aims of our funded research.
  • Pathology: Over 35 years of experience in a variety of morphological methods including 12 years as the Director of the Electron Microscopy Core at the University of Pennsylvania School of Medicine in a NIH funded Program Project in Reproductive Biology and over 10 years as Scientific Director of the Experimental Pathology and Histology/Immunocytochemistry facilities at USF Health and the Comprehensive Moffitt Cancer Center.
